TC Media Recipes
Current NCSU PTL Recipe (1 L)
1. Add 500 mL of diH2O to a >1000 mL beaker
2. Insert magnetic stir rod and place on stir plate at 125 speed
3. Add each item below individually and allow to dissolve before adding next ingredient:
2.3 g McCown Woody Plant Basal Salt Mixture
6 mL Thiamine (stock solution = 10 mg/ 100 mL)
1 mL 1000x MS vitamins
20 g Sucrose
0.1 g Myo-inositol
2 mL Zeatin (1 mg/ mL)
0.2 g Sequestrene
1.5 g MES
4. Adjust volume to 1 L
5. Calibrate pH meter
6. Bring pH up to 5
7. Add 4 g Agar
8. Autoclave on Liquid 20 setting (Should take 30-45 minutes)
9. Remove from autoclave and allow to cool for ~10 minutes
10. Pour media into Magenta boxes or culture tubes and allow to set and cool for ~1 hour or over night

- 1 ml MS vitamins

- pH 5
- 3.5 g agar

- 6 mL Thiamine
- 5 mL MS vitamins
- 30 g sucrose
- .1 g myo-Inositol
- 2 mg zeatin
- pH 5.2
- 6 g agar
