TC Solutions of Importance

1 mg/mL Zeatin Stock Solution

1. Add 100 mg of Zeatin to 100 mL volumetric flask or other glass container
2. Add 2-5 mL of 1M NaOH to dissolve the powder. Once completely dissolved, bring volume to 100 mL with molecular biological grade water. Note: Stirring to solution while adding water may be required to keep the material in solution.
3. Stock solutions may be stored at -20 degrees for up to 6 months
4. Add 1 mL of the stock solution to 1 L of medium to obtain a final concentration of 1 ug/mL of the zeatin in the culture medium

Dilute Bleach

1. 100 mL of 15% bleach solution
2. 15 mL bleach and 85 mL of water
3. For surface sterilization of plants add 3 drops of tween

Dilute Ethanol

1. 100 mL of 70% EtOH
2. 70 ML of 95% EtOH and 30 ML of distilled water

Dilute HCL

1. 225 mL of 1 M HCL
2. 18.75 mL of HCL and 206.25 mL water
3. Add acid to water

Dilute NaOH

1. 500 mL of 1 M=1 N NaOH
2. Dissolve 20 g NaOH in 500 mL water
